Adventures in Translocation: Studies of the Translocator Protein (TSPO) 18 kDa

نویسندگان

چکیده

The 18 kDa translocator protein (TSPO) is an evolutionarily conserved transmembrane found embedded in the outer mitochondrial membrane. A secondary target for benzodiazepine diazepam, TSPO has been a of interest researchers decades, particularly owing to its well-established links inflammatory conditions central and peripheral nervous systems. It become key biomarker assessing microglial activation using positron emission tomography (PET) imaging patients with diseases ranging from atherosclerosis Alzheimer’s disease. This Account describes research published by our group over past 15 years surrounding development ligands their use probing function this high-value target.

Stress sensitivity in patients with atopic dermatitis in relation to the translocator protein 18 kDa (TSPO).

Atopic dermatitis (AD) is a chronic inflammatory skin disease, characterized by pruritic and eczematous skin lesions and dermatitis that worsens under stressful conditions. However, the relation of these symptoms to an individual's stress sensitivity is not well understood. On the other hand, expression of the translocator protein (18 kDa) (TSPO), formerly known as the peripheral-type benzodiaz...

Regulation of Translocator Protein 18 kDa (TSPO) Expression in Rat and Human Male Germ Cells

Translocator protein 18 kDa (TSPO) is a high affinity cholesterol- and drug-binding protein highly expressed in steroidogenic cells, such as Leydig cells, where it plays a role in cholesterol mitochondrial transport. We have previously shown that TSPO is expressed in postnatal day 3 rat gonocytes, precursors of spermatogonial stem cells. Gonocytes undergo regulated phases of proliferation and m...

Translocator Protein 18 kDa (TSPO) Is Regulated in White and Brown Adipose Tissue by Obesity

Translocator protein 18 kDa (TSPO) is an outer-mitochondrial membrane transporter which has many functions including participation in the mitochondrial permeability transition pore, regulation of reactive oxygen species (ROS), production of cellular energy, and is the rate-limiting step in the uptake of cholesterol. TSPO expression is dysregulated during disease pathologies involving changes in...

The 18-kDa Translocator Protein (TSPO) Disrupts Mammary Epithelial Morphogenesis and Promotes Breast Cancer Cell Migration

Mitochondria play important roles in cancer progression and have emerged as viable targets for cancer therapy. Increasing levels of the outer mitochondrial membrane protein, 18-kDa translocator protein (TSPO), are associated with advancing breast cancer stage. In particular, higher TSPO levels are found in estrogen receptor (ER)-negative breast tumors, compared with ER-positive tumors. In this ...

A model radiopharmaceutical agent targeted to translocator protein 18 kDa (TSPO).

A stable Re complex containing an imidazopyridine ligand with a high affinity for TSPO has been synthesized as a model for new (99m)Tc or (188/186)Re-based radiopharmaceuticals to be used in SPECT diagnosis or in therapy, respectively. The new complex fac-[ReBr(CO)3(TZ6)], structurally characterized, showed high affinity (nanomolar concentration) for the target protein.
